      Effect of Resistance Training and Fish Protein Intake on Motor Unit Firing Pattern and Motor Function of Elderly

          Abstract

          We investigated the effect of resistance training and fish protein intake on the motor unit firing pattern and motor function in elderly. Fifty healthy elderly males and females (69.2 ± 4.7 years) underwent 6 weeks of intervention. We applied the leg-press exercise as resistance training and fish protein including Alaska pollack protein (APP) as nutritional supplementation. Subjects were divided into four groups: fish protein intake without resistance training (APP-CN, n = 13), placebo intake without resistance training (PLA-CN, n = 12), fish protein intake with resistance training (APP-RT, n = 12), and placebo intake with resistance training (PLA-RT, n = 13). Motor unit firing rates were calculated from multi-channel surface electromyography by the Convolution Kernel. For the chair-stand test, while significant increases were observed at 6 weeks compared with 0 week in all groups ( p < 0.05), significant increases from 0 to 3 weeks and 6 weeks were observed in APP-RT (18.2 ± 1.9 at 0 week to 19.8 ± 2.2 at 3 weeks and 21.2 ± 1.9 at 6 weeks) ( p < 0.05). Increase and/or decrease in the motor unit firing rate were mainly noted within motor units with a low-recruitment threshold in APP-RT and PLA-RT at 3 and 6 weeks (12.3 pps at 0 week to 13.6 pps at 3 weeks and 12.1 pps at 6 weeks for APP-RT and 12.9 pps at 0 week to 13.9 pps at 3 weeks and 14.1 pps at 6 weeks for PLA-RT at 50% of MVC) ( p < 0.05), but not in APP-CN or PLA-CN ( p > 0.05). Time courses of changes in the results of the chair-stand test and motor unit firing rate were different between APP-RT and PLA-RT. These findings suggest that, in the elderly, the effect of resistance training on the motor unit firing rate is observed in motor units with a low-recruitment threshold, and additional fish protein intake modifies these adaptations in motor unit firing patterns and the motor function following resistance training.

          Fundamentals of resistance training: progression and exercise prescription.

          Progression in resistance training is a dynamic process that requires an exercise prescription process, evaluation of training progress, and careful development of target goals. The process starts with the determination of individual needs and training goals. This involves decisions regarding questions as to what muscles must be trained, injury prevention sites, metabolic demands of target training goals, etc. The single workout must then be designed reflecting these targeted program goals including the choice of exercises, order of exercise, amount of rest used between sets and exercises, number of repetitions and sets used for each exercise, and the intensity of each exercise. For progression, these variables must then be varied over time and the exercise prescription altered to maintain or advance specific training goals and to avoid overtraining. A careful system of goal targeting, exercise testing, proper exercise technique, supervision, and optimal exercise prescription all contribute to the successful implementation of a resistance training program.

            Models of recruitment and rate coding organization in motor-unit pools.

            1. Isometric muscle force and the surface electromyogram (EMG) were simulated from a model that predicted recruitment and firing times in a pool of 120 motor units under different levels of excitatory drive. The EMG-force relationships that emerged from simulations using various schedules of recruitment and rate coding were compared with those observed experimentally to determine which of the modeled schemes were plausible representations of the actual organization in motor-unit pools. 2. The model was comprised of three elements: a motoneuron model, a motor-unit force model, and a model of the surface EMG. Input to the neuron model was an excitatory drive function representing the net synaptic input to motoneurons during voluntary muscle contractions. Recruitment thresholds were assigned such that many motoneurons had low thresholds and relatively few neurons had high thresholds. Motoneuron firing rate increased as a linear function of excitatory drive between recruitment threshold and peak firing rate levels. The sequence of discharge times for each motoneuron was simulated as a random renewal process. 3. Motor-unit twitch force was estimated as an impulse response of a critically damped, second-order system. Twitch amplitudes were assigned according to rank in the recruitment order, and twitch contraction times were inversely related to twitch amplitude. Nonlinear force-firing rate behavior was simulated by varying motor-unit force gain as a function of the instantaneous firing rate and the contraction time of the unit. The total force exerted by the muscle was computed as the sum of the motor-unit forces. 4. Motor-unit action potentials were simulated on the basis of estimates of the number and location of motor-unit muscle fibers and the propagation velocity of the fiber action potentials. The number of fibers innervated by each unit was assumed to be directly proportional to the twitch force. The area of muscle encompassing unit fibers was proportional to the number of fibers innervated, and the location of motor-unit territories were randomly assigned within the muscle cross section. Action-potential propagation velocities were estimated from an inverse function of contraction time. The train of discharge times predicted from the motoneuron model determined the occurrence of each motor-unit action potential. The surface EMG was synthesized as the sum of all motor-unit action-potential trains. 5.               Skeletal muscle adaptations during early phase of heavy-resistance training in men and women.

              An 8-wk progressive resistance training program for the lower extremity was performed twice a week to investigate the time course for skeletal muscle adaptations in men and women. Maximal dynamic strength was tested biweekly. Muscle biopsies were extracted at the beginning and every 2 wk of the study from resistance-trained and from nontrained (control) subjects. The muscle samples were analyzed for fiber type composition, cross-sectional area, and myosin heavy chain content. In addition, fasting blood samples were measured for resting serum levels of testosterone, cortisol, and growth hormone. With the exception of the leg press for women (after 2 wk of training) and leg extension for men (after 6 wk of training), absolute and relative maximal dynamic strength was significantly increased after 4 wk of training for all three exercises (squat, leg press, and leg extension) in both sexes. Resistance training also caused a significant decrease in the percentage of type IIb fibers after 2 wk in women and 4 wk in men, an increase in the resting levels of serum testosterone after 4 wk in men, and a decrease in cortisol after 6 wk in men. No significant changes occurred over time for any of the other measured parameters for either sex. These data suggest that skeletal muscle adaptations that may contribute to strength gains of the lower extremity are similar for men and women during the early phase of resistance training and, with the exception of changes in the fast fiber type composition, that they occur gradually.
