RESUMEN La estabilidad de un producto liofilizado es muy sensible al efecto de la humedad residual, por tal razón es importante controlarla en cada fase del proceso de liofilización, así como en la preparación del material de envase a utilizar. El presente trabajo tuvo como objetivo evaluar la influencia del tratamiento que reciben los tapones de bromobutilo en el contenido de humedad residual del inyectable liofilizado de omeprazol 40 mg. Materiales y Métodos: se muestrearon tres lotes de tapones de bromobutilo de 20 mm los cuales se esterilizaron por calor húmedo y se secaron. Se estimó el contenido de humedad de tres lotes industriales de omeprazol empleando tapones estériles y secos, durante doce meses, mediante la técnica de Karl Fischer, así como la valoración del inyectable por un método cromatográfico. Se demostró que el peso de los tapones esterilizados aumentó, como consecuencia de la absorción de humedad asociado a la esterilización. El secado de los tapones disminuyó el contenido de humedad inicial. Los lotes industriales del inyectable liofilizado de omeprazol, empleando tapones de bromobutilo estériles y secos, mantuvieron el contenido de humedad en el límite establecido, durante doce meses, así como la valoración, sin riesgos para la calidad del producto terminado.
ABSTRACT The stability of a lyophilized product is very sensitive to the effect of residual humidity, for that reason it is important not only to control it in every stage of the process of lyophilization, but also the preparation of the packaging material to use. The objective of the present work was to evaluate the influence of the treatment received by bromobutyl freeze drying closures on the residual moisture content of the lyophilized injectable omeprazole 40 mg. The three bromobutyl freeze-dried closures batches of 20 mm were tested, sterilized by moist heat and dried. The moisture content of three industrial batches of omeprazole was estimated using sterile and dried closures, for twelve months, by Karl Fischer technique, as well as the assay of the injectable by a chromatographic method. It were successfully demonstrated that the closure’s weight increased, as a result from the humidity absorption associated to the sterilization process. The drying process of the closures decreased the initial moisture content. The industrial batches of the freeze-dried injectable omeprazole, by using sterile and dry bromobutyl closures, kept the moisture content within the established limit, for twelve months, as well as the assay, without risks for the quality of the final product.
